Cross-Network PBMC Processing SOP
The Cross-Network PBMC Processing SOP provides instructions for processing PBMC at network site-affiliated laboratories, and is intended to simplify and clarify PBMC processing, especially at shared sites.
Proper viewing of the SOP and the PBMC SOP Processing Worksheet require the most recent version of Adobe Reader.
PBMC Processing Worksheet
For HVTN, the PBMC Processing Worksheet is required to track the timing of processing, calculations and documentation of problems that arise during processing. The worksheet is provided in Appendix A of the Cross-Network PBMC Processing SOP and below as a downloadable and editable form.
For ACTG, IMPAACT, HPTN and MTN, the use of a PBMC Processing Worksheet is recommended, but these networks may also use an equivalent record document, such as the Laboratory Data Management System (LDMS), for this purpose.
Note: For best results, follow these steps to use the worksheet:
- If you do not already have it, download the most recent version of Adobe Reader by clicking the icon above.
- Open the worksheet by clicking on the link below.
- Type the name of your lab into the fields at the top of each page.
- Save the file to your computer and print out the form for benchtop use.

Before using the SOP for the first time, thoroughly review all sections that apply to your network(s) for important notes and details. When you are familiar with the procedures, you may use the quick guides found in the appendices.
Reference Document
Also available is the Cross-Network PBMC Processing Reference Document, which includes information about PBMC processing at Center for HIV/AIDS Vaccine Immunology (CHAVI) laboratories.
Use of the reference SOP is not required, but you may find it a useful tool for reference and/or training.
